Treatment of the 6-chloropurine (I) with trimethylamine in DMF-THF produced the intermediate ammonium salt (II), which was displaced by KF in DMF at 80 C to afford fluoride (III). Finally, esterification of (III) with isobutyric anhydride in the presence of DMAP yielded the title isobutyrate ester.
